答えは①なのですがなぜ、of discount では行けないのかわかりません。
ofは12%の割引の、「の」にあたらないのですか？

にゃんさんのおっしゃる通り、意味を考えると、「なんで③ダメなん？」と思いますよね。
でも、英語の表現が間違っているのです。
① I(S) will give(V) you(IO) a twenty percent(形容詞) discount(名詞)(DO). ＜第4文型＞
他の例：It's a [ten(-)minute] walk from here. ここから歩いて10分の距離です。
※a [twenty percent] discount：ここでは[形容詞]として使っています。通常 twenty-percentと表現することが多いのですが。
③ I(S) will give(V) you(IO) ✕[a twenty percent](名詞)(IO) (of discount(名詞)(M) ＜第4文型＞
※a twenty percent：20%にaは付けられません。twenty percentは単数ではありません。

ofは20%の割引の、「の」にあたらないのですか？ →　あたりません。
twenty percent [of] discount → 割引[の]20% ／discount [of] 20 percent → 20%[の]割引
